Why the toxicologist behind the report matters
Every toxicology report is eventually read by someone whose job is to challenge it: an assessor reviewing a dossier, or a GMP inspector reviewing a cleaning validation. What makes it hold up is the science behind the derivation, the guideline it is written to, and the credentials of the toxicologist who signs it. Freyr’s board-certified toxicologists (DABT and ERT) prepare health-based exposure limits, impurity and E&L risk assessments, environmental risk assessments and F-value reports, each aligned with the applicable ICH, EMA or VICH guidance.

Preguntas frecuentes
01. What is regulatory toxicology?
Regulatory toxicology is the discipline of generating and interpreting safety data to meet the evidentiary requirements of medicines regulators. Its output is a defensible document (a risk assessment, an exposure limit, a justification) written to a named guideline and intended to withstand review by an assessor or GMP inspector.
02. Which ICH guidelines govern impurity toxicology?
Five apply directly. ICH Q3A(R2) covers impurities in new drug substances and Q3B(R2) in new drug products. Q3C(R8) covers residual solvents and Q3D(R2) elemental impurities. ICH M7(R2) governs mutagenic impurities and introduces the Threshold of Toxicological Concern approach. For veterinary medicinal products, VICH GL18 applies instead.
03. What qualifications should a regulatory toxicologist have?
Regulators expect the assessment to be performed by someone with demonstrable expertise in toxicology or pharmacology. Inspectors look for board certification (Diplomate of the American Board of Toxicology (DABT) or European Registered Toxicologist (ERT)) supported by a documented CV, and increasingly ask to see the assessor’s credentials during inspection.
04. What is an F-value report for child-resistant packaging?
An F-value is a toxicological threshold used to determine whether a medicinal product requires child-resistant packaging. It compares the amount of active substance a child could access in a single opening against the dose expected to cause harm. In the United States the requirement derives from 16 CFR 1700.20, with performance testing to ISO 8317.
05. When is an environmental risk assessment required for a medicinal product?
An ERA is required with every marketing authorization application in the EU, regardless of the outcome, under EMEA/CHMP/SWP/4447/00 Rev. 1. Phase I estimates the predicted environmental concentration; Phase II is triggered where that concentration exceeds the action limit or where the substance shows particular environmental properties.
06. Can one toxicology report be used across multiple regulatory markets?
Often yes, because the underlying ICH and EMA science is widely adopted. A PDE derived to EMA guidance is generally accepted by PIC/S member authorities and aligns with WHO TRS 1033. Differences arise in local expectations on format and route-specific limits, so reports intended for several markets should be scoped that way from the outset.
